Immunofluorescence (IF)/Immunocytochemistry (ICC)

(MBS9612254 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612254) and mouse anti-beta tubulin Ab(T0023) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI (blue).)

Related Product Information for anti-CPEB4 antibody
Function: Sequence-specific RNA-binding protein that binds to the cytoplasmic polyadenylation element (CPE), an uridine-rich sequence element (consensus sequence 5'-UUUUUAU-3') within the mRNA 3'-UTR (PubMed:24990967). RNA binding results in a clear conformational change analogous to the Venus fly trap mechanism (PubMed:24990967). Regulates activation of unfolded protein response (UPR) in the process of adaptation to ER stress in liver, by maintaining translation of CPE-regulated mRNAs in conditions in which global protein synthesis is inhibited (By similarity). Required for cell cycle progression, specifically for cytokinesis and chromosomal segregation (PubMed:26398195). Plays a role as an oncogene promoting tumor growth and progression by positively regulating translation of t-plasminogen activator/PLAT (PubMed:22138752). Stimulates proliferation of melanocytes (PubMed:27857118). In contrast to CPEB1 and CPEB3, does not play role in synaptic plasticity, learning and memory (By similarity).

Subcellular Location: Cytoplasm. Cell projection>Dendrite. Cell projection>Dendritic spine. Cell junction>Synapse>Postsynaptic density. Cell projection>Axon. Cell projection>Growth cone. Endoplasmic reticulum. Cytoplasm>Perinuclear region.

Tissue Specificity: Expressed in pancreas in islets and ductal cells (at protein level) (PubMed:22138752). Expressed in melanocytes (PubMed:27857118).

Subunit Structure: Interacts with TOB1.

Similarity: The 2 RRM domains and the C-terminal region mediate interaction with CPE-containing RNA (PubMed:24990967). The interdomain linker (564-579) acts as a hinge to fix the relative orientation of the 2 RRMs (PubMed:24990967). The ZZ domain (509-566) coordinates 2 Zn ions and is probably implicated in mediating interactions with other proteins in addition to increasing the affinity of the RRMs for the CPEs (By similarity). Unlike in CPEB1, a continuous polar interface is formed between the 2 RRMs (PubMed:24990967).Belongs to the RRM CPEB family.

Product Notes

The CPEB4 cpeb4 (Catalog #AAA9612254) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The CPEB4 Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig(100%), Bovine(100%), Horse(100%), Sheep(100%), Rabbit(100%), Dog(100%), Chicken(88%), Xenopus(100%) and may cross-react with other species as described in the data sheet. AAA Biotech's CPEB4 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). WB: 1:500-1:2000 IF: 1:100-1:500 ICC: 1:100-1:500 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the CPEB4 cpeb4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CPEB4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
