Function
Voltage-gated chloride channel (PubMed: 8112288, PubMed: 9122265, PubMed: 12456816). Plays an important role in membrane repolarization in skeletal muscle cells after muscle contraction. The CLC channel family contains both chloride channels and proton-coupled an ion transporters that exchange chloride or another anion for protons (Probable). The absence of conserved gating glutamate residues is typical for family members that function as channels (Probable).
Preparation and Storage
Maintain refrigerated at 2-8 degree C for up to 2 weeks.
For long term storage store at -20 degree C in small aliquots to prevent freeze-thaw cycles.

(Overlay histogram showing Hela cellsstained with (MBS9231248)(green line). The cellswere fixed with 2% paraformaldehyde (10min) and then permeabilized with 90%methanol for 10 min. The cells were thenicubated in 2% bovine serum albumin to block non-specific protein-protein interactionsfollowed by the antibody (MBS9231248, 1:25dilution) for 60 min at 37ºC. The secondaryantibody used was Goat-Anti-Rabbit IgG,DyLight® 488 Conjugated HighlyCross-Adsorbed(1583138) at 1/200 dilutionfor 40 min at 37ºC. Isotype control antibody(blue line) was rabbit IgG1 (1?g/1x10^6cells) used under the same conditions.Acquisition of >10, 000 events wasperformed.)

NCBI Description

The CLCN family of voltage-dependent chloride channel genes comprises nine members (CLCN1-7, Ka and Kb) which demonstrate quite diverse functional characteristics while sharing significant sequence homology. The protein encoded by this gene regulates the electric excitability of the skeletal muscle membrane. Mutations in this gene cause two forms of inherited human muscle disorders: recessive generalized myotonia congenita (Becker) and dominant myotonia (Thomsen). Alternative splicing results in multiple transcript variants. [provided by RefSeq, Mar 2012]
