Related Product Information for anti-CISH antibody
The suppressor of cytokine signaling (SOCS) family members are negative regulators of cytokine signal transduction that inhibit the Jak/Stat pathway. The SOCS family consists of at least 8 members including the originally identified cytokine-inducible SH2-containing protein (CIS1), as well as SOCS1-7. Each SOCS family member contains a central SH2 domain and a conserved carboxy-terminal motif designated as the SOCS box. These proteins are important regulators of cytokine signaling, proliferation, differentiation, and immune responses. CISH/CIS1, the first described member of the SOCS family, is induced by a number of cytokines including IL-2, IL-3, GM-CSF, and EPO. The CISH protein appears as a doublet around 32 and 37 kDa, the nature of which is unknown. CISH binds to phosphorylated cytokine receptors and can inhibit Stat5 activity. Expression of CISH is regulated by Stat5, thereby providing feedback modulation. Transgenic mice overexpressing CISH display phenotypes similar to Stat5 knockouts, including defects in mammary gland development and in T and NK cell regulation. Research studies have shown that polymorphisms within the CISH gene are associated with susceptibility to infectious diseases.

NCBI Description

The protein encoded by this gene contains a SH2 domain and a SOCS box domain. The protein thus belongs to the cytokine-induced STAT inhibitor (CIS), also known as suppressor of cytokine signaling (SOCS) or STAT-induced STAT inhibitor (SSI), protein family. CIS family members are known to be cytokine-inducible negative regulators of cytokine signaling. The expression of this gene can be induced by IL2, IL3, GM-CSF and EPO in hematopoietic cells. Proteasome-mediated degradation of this protein has been shown to be involved in the inactivation of the erythropoietin receptor. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Aug 2008]

Uniprot Description

CISH: SOCS family proteins form part of a classical negative feedback system that regulates cytokine signal transduction. CIS is involved in the negative regulation of cytokines that signal through the JAK-STAT5 pathway such as erythropoietin, prolactin and interleukin 3 (IL3) receptor. Inhibits STAT5 trans-activation by suppressing its tyrosine phosphorylation. May be a substrate- recognition component of a SCF-like ECS (Elongin BC-CUL2/5-SOCS- box protein) E3 ubiquitin-protein ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins. Stably associated with the tyrosine-phosphorylated IL3 receptor beta chain and tyrosine-phosphorylated EPO receptor (EPOR). By a subset of cytokines including EPO/erythropoietin. Expressed in various epithelial tissues. Abundantly expressed in liver and kidney, and to a lesser extent in lung. The tissue distribution of isoforms 1 and 1B is distinct. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The CISH cish (Catalog #AAA3005500) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The CISH polyclonal antibody reacts with Rat, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's CISH can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC). WB: 1:500~1:1000 IHC: 1:50~1:200. Researchers should empirically determine the suitability of the CISH cish for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CISH, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
