Rabbit anti-Human CDK4 Polyclonal Antibody | anti-CDK4 antibody
CDK4 Antibody (C-term)
WB~~1:1000
Immunofluorescence (IF)
(Fluorescent image of Hela cell stained with CDK4 Antibody (C-term). Hela cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with CDK4 primary antibody (1:25, 1 h at 37 degree). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:400, 50 min at 37 degree).Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37 degree).CDK4 immunoreactivity is localized to Cytoplasm significantly.)
Western Blot (WB)
(Western blot analysis of anti-CDK4 Pab in HL-60 cell lysate. CDK4 (arrow) was detected using purified Pab. Secondary HRP-anti-rabbit was used for signal visualization with chemiluminescence.)
Western Blot (WB)
(Western blot analysis of CDK4 (arrow) using rabbit polyclonal CDK4 Antibody (C-term). 293 cell lysates (2 ug/lane) either nontransfected (Lane 1) or transiently transfected with the CDK4 gene (Lane 2) (Origene Technologies).)
Immunohistochemistry (IHC)
(Formalin-fixed and paraffin-embedded human cancer tissue reacted with the primary antibody, which was peroxidase-conjugated to the secondary antibody, followed by AEC staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated. BC = breast carcinoma; HC = hepatocarcinoma.)
Immunohistochemistry (IHC)
(Formalin-fixed and paraffin-embedded human prostate carcinoma with CDK4 Antibody (C-term), which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated.)
Flow Cytometry (FC/FACS)
(CDK4 Antibody (C-term) flow cytometric analysis of HL60 cells (bottom histogram) compared to a negative control cell (top histogram).FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.)

NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a member of the Ser/Thr protein kinase family. This protein is highly similar to the gene products of S. cerevisiae cdc28 and S. pombe cdc2. It is a catalytic subunit of the protein kinase complex that is important for cell cycle G1 phase progression. The activity of this kinase is restricted to the G1-S phase, which is controlled by the regulatory subunits D-type cyclins and CDK inhibitor p16(INK4a). This kinase was shown to be responsible for the phosphorylation of retinoblastoma gene product (Rb). Mutations in this gene as well as in its related proteins including D-type cyclins, p16(INK4a) and Rb were all found to be associated with tumorigenesis of a variety of cancers. Multiple polyadenylation sites of this gene have been reported. [provided by RefSeq, Jul 2008]
Uniprot Description
CDK4: a protein kinase of the CDK family that is important for cell cycle G1 phase progression. Its activity is restricted to the G1-S phase. Controlled by the regulatory subunits D-type cyclins and CDK inhibitor p16(INK4a). Phosphorylates the retinoblastoma gene product (Rb). Point mutations found in somatic and familial melanoma. Amplified in sarcomas, glioma and lymphoma. Amplified, methylated or deleted in head and neck squamous cell carcinoma. Overexpression drives epithelial tumors in mice. Disruption makes mice resistant to cancer. Inhibitor: PD332991.

Product Notes
The CDK4 cdk4 (Catalog #AAA9210247) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 273-305. The CDK4 Antibody (C-term) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's CDK4 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), ELISA (EIA), Immunohistochemistry (IHC), Western Blot (WB), Immunofluorescence (IF). IF~~1:10~50 WB~~1:1000. Researchers should empirically determine the suitability of the CDK4 cdk4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CDK4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
