Preparation and Storage

CD34 (Hematopoietic progenitor cell antigen CD34) is a monomeric cell surface antigen with a molecular mass of approximately 110 kD that is selectively expressed on human hematopoietic progenitor cells. The CD34 protein is a member of a family of single-pass transmembrane sialomucin proteins that show expression on early hematopoietic and vascular-associated tissue.1 It plays an essential role in early hematopoiesis.2 CD34 is also an important adhesion molecule and is required for T cells to enter lymph nodes. It is expressed on lymph node endothelia whereas the L-selectin to which it binds is on the T cell.3,4

The protein encoded by this gene may play a role in the attachment of stem cells to the bone marrow extracellular matrix or to stromal cells. This single-pass membrane protein is highly glycosylated and phosphorylated by protein kinase C. Two transcript variants encoding different isoforms have been found for this gene. Function: Possible adhesion molecule with a role in early hematopoiesis by mediating the attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells. Could act as a scaffold for the attachment of lineage specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components. Presents carbohydrate ligands to selectins.

Subcellular location: Membrane; Single-pass type I membrane protein.

Tissue specificity: Selectively expressed on hematopoietic progenitor cells and the small vessel endothelium of a variety of tissues.

Developmental stage: On early hematopoietic progenitor cells.

Post-translational modification: Highly glycosylated.Phosphorylated on serine residues by PKC. Ref.10

Sequence similarities: Belongs to the CD34 family.

Sequence caution: The sequence AAA03181.1 differs from that shown. Reason: Erroneous initiation.
