Western Blot (WB)
(Western blot analysis of CD33 expression in SKOV3 whole cell lysates (lane 1). CD33 at 45KD; 67KD was detected using rabbit anti-CD33 Antigen Affinity purified polyclonal antibody at 0.5 ug/mL. The blot was developed using chemiluminescence (ECL) method. )
Immunohistochemistry (IHC)
(CD33 was detected in paraffin-embedded sections of human tonsil tissues using rabbit anti- CD33 Antigen Affinity purified polyclonal antibody at 1ug/mL. The immunohistochemical section was developed using SABC method. )
Flow Cytometry (FC/FACS)
(Figure 3. Flow Cytometry analysis of U937 cells using anti-CD33 antibody (MBS178432).Overlay histogram showing U937 cells stained with MBS178432 (Blue line). The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-CD33 Antibody (MBS178432,1ug/1x10^6 cells) for 30 min at 20 degree C. DyLight 488 conjugated goat anti-rabbit IgG (5-10ug/1x10^6 cells) was used as secondary antibody for 30 minutes at 20 degree C. Isotype control antibody (Green line) was rabbit IgG (1ug/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control. )

NCBI and Uniprot Product Information
Uniprot Description
CD33: Putative adhesion molecule of myelomonocytic-derived cells that mediates sialic-acid dependent binding to cells. Preferentially binds to alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. In the immune response, may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatase(s) via their SH2 domain(s) that block signal transduction through dephosphorylation of signaling molecules. Induces apoptosis in acute myeloid leukemia (in vitro). Belongs to the immunoglobulin superfamily. SIGLEC (sialic acid binding Ig-like lectin) family.
Protein type: Membrane protein, integral; Cell adhesion
Chromosomal Location of Human Ortholog: 19q13.3
Cellular Component: external side of plasma membrane; integral to plasma membrane; plasma membrane
Molecular Function: protein binding; receptor activity
Biological Process: cell-cell signaling; negative regulation of cell proliferation; regulation of immune response; signal transduction

The CD33 cd33 (Catalog #AAA178432) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-CD33 Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's CD33 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC) Paraffin/Frozen, Immunocytochemistry (ICC), Flow Cytometry (FC/FACS). Western Blot: 0.1-0.5ug/ml Immunohistochemistry - Paraffin: 0.5-1ug/ml; Antigen Retrieval: By Heat Immunohistochemistry - Frozen: 0.5-1ug/ml Immunocytochemistry: 0.5-1ug/ml Flow Cytometry: 1-3ug/1x106 cells Tested Species: In-house tested species with positive results. By Heat: Boiling the paraffin sections in 10mM citrate buffer, pH6.0, for 20mins is required for the staining of formalin/paraffin sections. Other applications have not been tested. Researchers should empirically determine the suitability of the CD33 cd33 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD33, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
