Host Note
Polyclonal CD20 antibody was produced from sera of rabbits immunized with synthetic protein containing CD20 antigenic determinants.
Reconstitution
A quick spin of the vial followed by reconstitution in 0.01M PBS pH 7.4 or other diluents.
Preparation and Storage
The lyophilized antibody is stable for at least 1 year from date of receipt at -20 degree C.
Upon reconstitution, this antibody can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months without detectable loss of activity.

Avoid repeated freeze/thaw cycles.
Shelf Life: >12 months

CD20 is a signature B-cell differentiation antigen strongly expressed on the surface of mature normal as well as malignant B cells. It is also expressed, but at lower levels and with larger variance, on more immature B cells and their malignant counterparts found in B-cell precursor (BCP) acute lymphoblastic leukemia. The precise function of CD20 remains unknown, it appears to act as part of an ion channel expressed in the plasma membrane,recently shown that CD20 may operate as a store operated calcium (Ca2+) channel facilitating entry of extracellular Ca2+ following BCR-induced emptying of intracellular stores. Interestingly, despite this presumably important role, CD20- deficient mice are without obvious phenotype, which suggests that either this function is not critical for B cell physiology or that molecular redundancy exists.

NCBI Description

This gene encodes a member of the membrane-spanning 4A gene family. Members of this nascent protein family are characterized by common structural features and similar intron/exon splice boundaries and display unique expression patterns among hematopoietic cells and nonlymphoid tissues. This gene encodes a B-lymphocyte surface molecule which plays a role in the development and differentiation of B-cells into plasma cells. This family member is localized to 11q12, among a cluster of family members. Alternative splicing of this gene results in two transcript variants which encode the same protein. MS4A1: This protein may be involved in the regulation of B-cell activation and proliferation. Defects in MS4A1 are the cause of immunodeficiency common variable type 5 (CVID5); also called antibody deficiency due to CD20 defect. CVID5 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B-cells is usually in the normal range, but can be low. Belongs to the MS4A family.
