Immunohistochemistry (IHC)

(Immunohistochemical analysis of CARD11 staining in human brain formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.)

(Immunofluorescent analysis of CARD11 staining in K562 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).)

NCBI Description

The protein encoded by this gene belongs to the membrane-associated guanylate kinase (MAGUK) family, a class of proteins that functions as molecular scaffolds for the assembly of multiprotein complexes at specialized regions of the plasma membrane. This protein is also a member of the CARD protein family, which is defined by carrying a characteristic caspase-associated recruitment domain (CARD). This protein has a domain structure similar to that of CARD14 protein. The CARD domains of both proteins have been shown to specifically interact with BCL10, a protein known to function as a positive regulator of cell apoptosis and NF-kappaB activation. When expressed in cells, this protein activated NF-kappaB and induced the phosphorylation of BCL10. [provided by RefSeq, Jul 2008]

Uniprot Description

CARD11: a CARD (caspase recruitment domain) protein that belongs to the membrane-associated guanylate kinase (MAGUK) family. MAGUK proteins function as molecular scaffolds for the assembly of multiprotein complexes at specialized regions of the plasma membrane. Contains an N-terminal CARD domain, a central coiled-coil domain, and a C-terminal region containing a PDZ, an SH3, and a GUK. Its CARD domain associates with the CARD domain of BCL10. BCL10 activates NF-kappaB through the IkappaB kinase complex in response to upstream stimuli. Apparently activates NF-kappaB and induce the phosphorylation of BCL10. Detected in adult peripheral blood leukocytes, thymus, spleen and liver. Also found in promyelocytic leukemia HL-60 cells, chronic myelogenous leukemia K562 cells, Burkitt's lymphoma Raji cells and colorectal adenocarcinoma SW480 cells.

Product Notes

The CARD11 card11 (Catalog #AAA8242680) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-CARD11 Antibody reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's CARD11 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC), Immunofluorescence (IF), Immunocytochemistry (ICC). Western Blot: (1/500 - 1/1000); Immunohistochemistry: (1/100 - 1/200); Immunofluorescence/Immunocytochemistry: (1/100 - 1/500). Researchers should empirically determine the suitability of the CARD11 card11 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CARD11, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
