Isotype
IgG
Specificity
This item recognises human Carabin, also known as TBC1 domain family member 10C (TBC1D10C), a 50 kDa protein belonging to the TBC1 domain family of proteins.

Carabin differs from other members of the TBC1 domain protein family, which appear to play a role in the regulation of cell growth and differentiation. Studies demonstrate that Carabin is part of a negative regulatory loop for the intracellular TCR signalling pathway and acts as an inhibitor of the Ras signalling pathway. Carabin is also believed to mediate crosstalk between calcineurin and Ras.

Carabin is most abundantly expressed in spleen and peripheral blood leucocytes.

This item does not cross react with either TBC1D10A or TBC1D10B.
Form/Format
Purified
Purified IgG - liquid
Concentration
IgG concentration 1.0mg/ml (varies by lot)
Sequence Length
310
Applicable Applications for anti-TBC1D10C antibody
Immunohistology Paraffin*, Western Blot (WB)
Application Notes
Western Blot: This item detects a band of approximately 55kDa in human spleen tissue cell lysates.
Immunohistology - Paraffin: Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose.
Western Blotting: Minimum Dilution: 1; Maximum Dilution: 2ug/ml
Perservative Stabilisers
0.02% Sodium Azide (NaN3)
Immunogen
A 17 amino acid peptide from near the amino terminus of human Carabin.
Buffer Solution
Phosphate buffered saline
Antiserum Preparation
Antiserum to human Carabin was raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.
Target Species
Human
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 18 months from date of despatch.

NCBI Description

The protein encoded by this gene has an N-terminal Rab-GTPase domain and a binding site at the C-terminus for calcineurin, and is an inhibitor of both the Ras signaling pathway and calcineurin, a phosphatase regulated by calcium and calmodulin. Genes encoding similar proteins are located on chromosomes 16 and 22. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jan 2013]

Uniprot Description

TBC1D10C: Inhibits the Ras signaling pathway through its intrinsic Ras GTPase-activating protein (GAP) activity. Acts as a negative feedback inhibitor of the calcineurin signaling pathway that also mediates crosstalk between calcineurin and Ras.

Protein type: GAPs; GAPs, Rab

Chromosomal Location of Human Ortholog: 11q13.2

Cellular Component: filopodium membrane; membrane; plasma membrane

Biological Process: negative regulation of B cell activation; negative regulation of NFAT protein import into nucleus; retrograde transport, endosome to Golgi

Product Notes

The TBC1D10C tbc1d10c (Catalog #AAA222937) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI CARABIN (N-TERMINAL) reacts with Mouse N.B. Antibody reactivity and working conditions may vary between species. and may cross-react with other species as described in the data sheet. AAA Biotech's CARABIN can be used in a range of immunoassay formats including, but not limited to, Immunohistology Paraffin*, Western Blot (WB). Western Blotting: Minimum Dilution: 1; Maximum Dilution: 2ug/ml. Researchers should empirically determine the suitability of the TBC1D10C tbc1d10c for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CARABIN, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
