Application Notes
AXIN2 antibody can be used for detection of AXIN2 by Western blot at 1 ug/mL. Antibody can also be used for immunohistochemistry starting at 5 ug/mL. For immunofluorescence start at 20 ug/mL. Antibody validated: Western Blot in mouse samples; Immunohistochemistry in mouse and rat samples and Immunofluorescence in mouse and rat samples.

All other applications and species not yet tested.
Optimal dilutions for each application to be determined by the researcher.

Preparation and Storage
AXIN2 antibody can be stored at 4 degree C for three months and -20 degree C, stable for up to one year. As with all antibodies care should be taken to avoid repeated freeze thaw cycles. Antibodies should not be exposed to prolonged high temperatures.

AXIN2 Antibody: Like the related protein AXIN1, AXIN2 is thought to play an important role in the regulation of the stability of beta-catenin in the Wnt signaling pathway. In mouse, AXIN2 organizes a multiprotein complex of APC (adenomatous polyposis of the colon), beta-catenin, glycogen synthase kinase 3-beta, and AXIN2, which leads to the degradation of beta-catenin, which is thought to be an important event in the genesis of a number of malignancies. The AXIN2 gene has been mapped to 17q23-q24, a region that shows frequent loss of heterozygosity in breast cancer, neuroblastoma, and other tumors. Mutations in this gene have been associated with colorectal cancer with defective mismatch repair.

NCBI Description

The Axin-related protein, Axin2, presumably plays an important role in the regulation of the stability of beta-catenin in the Wnt signaling pathway, like its rodent homologs, mouse conductin/rat axil. In mouse, conductin organizes a multiprotein complex of APC (adenomatous polyposis of the colon), beta-catenin, glycogen synthase kinase 3-beta, and conductin, which leads to the degradation of beta-catenin. Apparently, the deregulation of beta-catenin is an important event in the genesis of a number of malignancies. The AXIN2 gene has been mapped to 17q23-q24, a region that shows frequent loss of heterozygosity in breast cancer, neuroblastoma, and other tumors. axin 2: is a negative regulator of the Wnt pathway, which is critical in stem cell signaling, morphogenesis, the mesenchymal-epithelial transition, and many cancers. Axin functions as a tumor suppressor. Probably facilitates the phosphorylation of beta-catenin and APC by GSK3B, leading to their ubiquitination and subsequent proteolysis. Is downregulated during progression of esophageal squamous cell carcinoma. Axin1/2 mediate cross-talk between TGF-beta and Wnt signaling pathways.
