Related Product Information for anti-ASIP antibody
Several common diseases such as type II diabetes, hypertension, cardiovascular diseases, hyperlipidemia, and some cancers are associated with obesity. An abnormal increase in body fat relative to lean tissue mass has been used as an indicator of obesity. High fat diet, certain environmental factors, and genetic linkage are the primary causes of obesity. In order to understand the genetic basis of obesity, several monogenic murine obesity models have been characterized including obese (Ob), diabetes (db), fat (fat), agouti yellow (Ay), and tubby (tub). More recently, Tub, the human homolog of mouse Tub, TULP1 & TULP2 (for Tubby Like Proteins) and Agouti related protein (AGRP) have been cloned. The obesity associated with Ay mice may be due to ectopic expression of a secreted protein Agouti. Agouti protein (132aa in human) is normally expressed in skin. Its ubiquitous expression, such as in rare allele lethal yellow, causes yellow coat., linear growth, immune defects and obesity. Agouti is a paracrine-signaling molecule that affects pigmentation by inhibiting the melanocortin receptor 1 (MCR-1 and possibly MC2R and MC4R).

NCBI Description

In mice, the agouti gene encodes a paracrine signaling molecule that causes hair follicle melanocytes to synthesize pheomelanin, a yellow pigment, instead of the black or brown pigment, eumelanin. Pleiotropic effects of constitutive expression of the mouse gene include adult-onset obesity, increased tumor susceptibility, and premature infertility. This gene is highly similar to the mouse gene and encodes a secreted protein that may (1) affect the quality of hair pigmentation, (2) act as a pharmacological antagonist of alpha-melanocyte-stimulating hormone, (3) play a role in neuroendocrine aspects of melanocortin action, and (4) have a functional role in regulating lipid metabolism in adipocytes. [provided by RefSeq, Jul 2008]

Uniprot Description

ASIP: Involved in the regulation of melanogenesis. The binding of ASP to MC1R precludes alpha-MSH initiated signaling and thus blocks production of cAMP, leading to a down-regulation of eumelanogenesis (brown/black pigment) and thus increasing synthesis of pheomelanin (yellow/red pigment). In higher primates, agouti may affect the quality of hair pigmentation rather than its pattern of deposition. Could well play a role in neuroendocrine aspects of melanocortin action. May have some functional role in regulating the lipid metabolism with adipocytes.

Product Notes

The ASIP asip (Catalog #AAA6008951) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Agouti reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Agouti can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC), Immunofluorescence (IF). Suitable for use in Immunohistochemistry and Immunofluorescence. Dilution: Immunofluorescence: 1:200 Immunohistochemistry (PAP or ABC): 1:500. Researchers should empirically determine the suitability of the ASIP asip for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: His-Leu-Pr o-Pro-Glu- Glu-Lys-Le u-Arg-Asp- Asp-Arg-Se r-Leu-Arg- Ser-Asn-Se r-Ser-Val- Asn-Leu-Le u-Asp-Val- Pro-Ser-Va l-Ser-Ile- Val-Ala-Le u-Asn-Lys- Lys-Ser-Ly s-Gln-Ile- NH2. It is sometimes possible for the material contained within the vial of "Agouti, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
