Materials Provided
1. Capture Antibody 0.3 mg/mL of mouse anti-PLXDC1 monoclonal antibody. Dilute to a working concentration of 2ug/mL in CBS before coating.
2. Detection Antibody 0.5 mg/mL mouse anti-PLXDC1 monoclonal antibody conjugated to horseradish-peroxidase (HRP). Dilute to working concentration of 0.5ug/mL in detection antibody dilution buffer before use.
3. Standard Each vial contains 270 ng of recombinant PLXDC1. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20 degree C to -80 degree C in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilution buffer, and a high standard of 10ng/mL is recommended.

Preparation and Storage

Detection Antibody: Protect it from prolonged exposure to light. Aliquot and store at -20 degree C to -80 degree C and for up to 6 months from date of receipt. Avoid repeated freeze-thaw cycles.
Standard: Store lyophilized standard at -20 degree C to -80 degree C for up to 6 months from date of receipt. Aliquot and store the reconstituted Standard at -80 degree C for up to 1 month. Avoid repeated freeze-thaw cycles.

Plexin domain-containing protein 1, also known as tumor endothelial marker 3, tumor endothelial marker 7 and PLXDC1 and TEM3, is a secreted, cytoplasm and single-pass type I membrane protein that belongs to the plexin family. PLXDC1/TEM3 is detected in endothelial cells from colorectal cancer, and in endothelial cells from primary cancers of the lung, liver, pancreas, breast and brain. It is expressed in fibrovascular membrane with increased expression in individuals with proliferative diabetic retinopathy. PLXDC1/TEM3 is not detectable in endothelial cells from normal tissue. PLXDC1/TEM3 plays a critical role in endothelial cell capillary morphogenesis. PLXDC1/TEM3 may play a significant role in the proliferation and maintenance of neovascular endothelial cells in the formation of fibrovascular membranes (FVMs).
