Related Product Information for POMC peptide
Description: Pro-opiomelanocortin (POMC) is a precursor polypeptide with 241 amino acid residues. It is synthesized by corticotrope cells of the anterior pituitary gland, melanotrope cells of the intermediate lobe of the pituitary gland, about 3000 neurons in the arcuate nucleus of the hypothalamus, and smaller populations of neurons in the dorsomedial hypothalamus and brainstem. adrenocorticotrophic hormone (ACTH) and gamma LPH in the anterior pituitary gland CLIP, gamma LPH, gamma MSH and beta-endorphin in the intermediate lobe gamma-MSH met-enkephalin Each of these peptides is packaged in large dense-core vesicles that are released from the cells by exocytosis in response to appropriate stimulation. gamma MSH produced by neurons in the arcuate nucleus has important roles in the regulation of appetite and sexual behavior, while ?-MSH secreted from the intermediate lobe of the pituitary regulates the production of melanin. ACTH is a peptide hormone that regulates the secretion of glucocorticoids from the adrenal cortex. beta-endorphin and met-enkephalin are endogenous opioid peptides with widespread actions in the brain.

NCBI Description

This gene encodes a polypeptide hormone precursor that undergoes extensive, tissue-specific, post-translational processing via cleavage by subtilisin-like enzymes known as prohormone convertases. There are eight potential cleavage sites within the polypeptide precursor and, depending on tissue type and the available convertases, processing may yield as many as ten biologically active peptides involved in diverse cellular functions. The encoded protein is synthesized mainly in corticotroph cells of the anterior pituitary where four cleavage sites are used; adrenocorticotrophin, essential for normal steroidogenesis and the maintenance of normal adrenal weight, and lipotropin beta are the major end products. In other tissues, including the hypothalamus, placenta, and epithelium, all cleavage sites may be used, giving rise to peptides with roles in pain and energy homeostasis, melanocyte stimulation, and immune modulation. These include several distinct melanotropins, lipotropins, and endorphins that are contained within the adrenocorticotrophin and beta-lipotropin peptides. Mutations in this gene have been associated with early onset obesity, adrenal insufficiency, and red hair pigmentation. Alternatively spliced transcript variants encoding the same protein have been described. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: ACTH stimulates the adrenal glands to release cortisol.MSH (melanocyte-stimulating hormone) increases the pigmentation of skin by increasing melanin production in melanocytes.Beta-endorphin and Met-enkephalin are endogenous opiates.

Subcellular location: Secreted.

Tissue specificity: ACTH and MSH are produced by the pituitary gland.

Post-translational modification: Specific enzymatic cleavages at paired basic residues yield the different active peptides.O-glycosylated; reducing sugar is probably N-acetylgalactosamine.

Involvement in disease: Obesity (OBESITY) [MIM:601665]: A condition characterized by an increase of body weight beyond the limitation of skeletal and physical requirements, as the result of excessive accumulation of body fat.Note: Disease susceptibility may be associated with variations affecting the gene represented in this entry. Ref.30Pro-opiomelanocortinin deficiency (POMCD) [MIM:609734]: Affected individuals present early-onset obesity, adrenal insufficiency and red hair.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.24

Sequence similarities: Belongs to the POMC family.

Product Notes

The POMC pomc (Catalog #AAA659146) is a Peptide and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: Synthetic peptide to porcine POMC Precursor (aa27-52) (linear) Trp-Cys-Le u-Glu-Ser- Ser-Gln-Cy s-Gln-Asp- Leu-Ser-Th r-Glu-Ser- Asn-Leu-Le u-Ala-Cys- Ile-Arg-Al a-Cys-Lys- Pro. It is sometimes possible for the material contained within the vial of "Proopiomelanocortin, Peptide" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
