Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate OPRD1 in samples. An antibody specific for OPRD1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyOPRD1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for OPRD1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of OPRD1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description
G-protein coupled receptor that functions as receptor for endogenous enkephalins and for a subset of other opioids. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of down-stream effectors, such as adenylate cyclase. Signaling leads to the inhibition of adenylate cyclase activity. Inhibits neurotransmitter release by reducing calcium ion currents and increasing potassium ion conductance. Plays a role in the perception of pain and in opiate-mediated analgesia. Plays a role in developing analgesic tolerance to morphine.
