Principle of the Assay: NT-3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-NT-3 antibody and an NT-3-HRP conjugate. The assay sample and buffer are incubated together with NT-3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NT-3 concentration since NT-3 from samples and NT-3-HRP conjugate compete for the anti-NT-3 antibody binding site. Since the number of sites is limited, as more sites are occupied by NT-3 from the sample, fewer sites are left to bind NT-3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NT-3 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the neurotrophin family, that controls survival and differentiation of mammalian neurons. This protein is closely related to both nerve growth factor and brain-derived neurotrophic factor. It may be involved in the maintenance of the adult nervous system, and may affect development of neurons in the embryo when it is expressed in human placenta. NTF3-deficient mice generated by gene targeting display severe movement defects of the limbs. The mature peptide of this protein is identical in all mammals examined including human, pig, rat and mouse. [provided by RefSeq, Jul 2008]
