Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to Pro-NRG1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Pro-NRG1 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Pro-NRG1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Pro-NRG1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Uniprot Description

Direct ligand for the ERBB tyrosine kinase receptors. The multiple isoforms perform diverse functions: cysteine-rich domain containing isoforms (isoform 2-isoform 4) probably regulate the expression of nicotinic acetylcholine receptors at developing interneuronal synapses. Isoform Ig-NRG is required for the initial induction and/or maintenance of the mature levels of acetylcholine receptors at neuromuscular synapses (PubMed:8453670, PubMed:9491987). Binds to ERBB3 and integrins to form a complex which is essential for NRG1-ERBB signaling ().
