Intended Uses: This ELISA kit applies to the invitro quantitative determination of HumanNFATC2concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to NFATC2. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for NFATC2and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain NFATC2, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of NFATC2.You can calculate the concentration of NFATC2in the samples by comparing the OD of the samples to the standard curve.

NCBI Description

This gene is a member of the nuclear factor of activated T cells (NFAT) family. The product of this gene is a DNA-binding protein with a REL-homology region (RHR) and an NFAT-homology region (NHR). This protein is present in the cytosol and only translocates to the nucleus upon T cell receptor (TCR) stimulation, where it becomes a member of the nuclear factors of activated T cells transcription complex. This complex plays a central role in inducing gene transcription during the immune response. Alternate transcriptional splice variants encoding different isoforms have been characterized. Uniprot Description

NFAT1: a transcription factor that plays a role in the inducible expression of cytokine genes in T cells, especially in the induction of the IL-2, IL-3, IL-4, TNF-alpha and GM-CSF. It is present in a phosphorylated the cytosol of unstimulated T cells. After T-cell activation, it is dephosphorylated by calcineurin and translocates to the nucleus as a part of a transcription complex. Two splice variant isoforms have been identified.
