Principle of the Assay: N-ACHR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-N-ACHR antibody and an N-ACHR-HRP conjugate. The assay sample and buffer are incubated together with N-ACHR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the N-ACHR concentration since N-ACHR from samples and N-ACHR-HRP conjugate compete for the anti-N-ACHR antibody binding site. Since the number of sites is limited, as more sites are occupied by N-ACHR from the sample, fewer sites are left to bind N-ACHR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The N-ACHR concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a nicotinic acetylcholine receptor, which belongs to a superfamily of ligand-gated ion channels that play a role in fast signal transmission at synapses. These pentameric receptors can bind acetylcholine, which causes an extensive change in conformation that leads to the opening of an ion-conducting channel across the plasma membrane. This protein is an integral membrane receptor subunit that can interact with either nAChR beta-2 or nAChR beta-4 to form a functional receptor. Mutations in this gene cause nocturnal frontal lobe epilepsy type 1. Polymorphisms in this gene that provide protection against nicotine addiction have been described.
