Principle of the assay: MPI ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MPI antibody and an MPI-HRP conjugate. The assay sample and buffer are incubated together with MPI-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MPI concentration since MPI from samples and MPI-HRP conjugate compete for the anti-MPI antibody binding site. Since the number of sites is limited, as more sites are occupied by MPI from the sample, fewer sites are left to bind MPI-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MPI concentration in each sample is interpolated from this standard curve.

Phosphomannose isomerase catalyzes the interconversion of fructose-6-phosphate and mannose-6-phosphate and plays a critical role in maintaining the supply of D-mannose derivatives, which are required for most glycosylation reactions. Mutations in the MPI gene were found in patients with carbohydrate-deficient glycoprotein syndrome, type Ib. Alternative splicing results in multiple transcript variants. Defects in MPI are the cause of congenital disorder of glycosylation type 1B (CDG1B); also known as carbohydrate-deficient glycoprotein syndrome type Ib (CDGS1B). Congenital disorders of glycosylation are metabolic deficiencies in glycoprotein biosynthesis that usually cause severe mental and psychomotor retardation.
