Test Principle: This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to OX has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled OX and unlabeled OX (Standards or samples) with the pre-coated antibody specific to OX. After incubation the unbound conjugate is washed off. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of OX in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of OX in the sample.

NCBI Description

This gene encodes a hypothalamic neuropeptide precursor protein that gives rise to two mature neuropeptides, orexin A and orexin B, by proteolytic processing. Orexin A and orexin B, which bind to orphan G-protein coupled receptors Hcrtr1 and Hcrtr2, function in the regulation of sleep and arousal. This neuropeptide arrangement may also play a role in feeding behavior, metabolism, and homeostasis. [provided by RefSeq, Sep 2015]

Neuropeptides that play a significant role in the regulation of food intake and sleep-wakefulness, possibly by coordinating the complex behavioral and physiologic responses of these complementary homeostatic functions. Orexin-A binds to both OX1R and OX2R with a high affinity, whereas orexin-B binds only to OX2R with a similar high affinity.
