Preparation and Storage
For unopened kits: All the reagents should be kept at 4 degree C upon receipt.
For opened kits: Once the kit is opened, the remaining reagents still need to be stored according to the above storage conditions. In addition, return the unused wells to the foil pouch containing the desiccant pack and reseal along entire edge of zip-seal.

Test Principle: The microtiter plate provided in this kit has been pre-coated with an antibody specific to GRIA3. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific to GRIA3. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain GRIA3, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of GRIA3 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

This gene encodes a multi-pass transmembrane protein that forms a homotetramer or heterotetramer in neuronal cells. The encoded protein is a ligand-gated ion channel that responds to the neurotransmitter L-glutamate to promote synaptic transmission. Deficiency of this gene leads to behavioral phenotypes. The transcript is subject to RNA editing at codon 769 (AGA->GGA; R->G). Alternative splicing results in multiple transcript variants. [provided by RefSeq, Aug 2013]

Uniprot Description

Receptor for glutamate that functions as ligand-gated ion channel in the central nervous system and plays an important role in excitatory synaptic transmission. L-glutamate acts as an excitatory neurotransmitter at many synapses in the central nervous system. Binding of the excitatory neurotransmitter L-glutamate induces a conformation change, leading to the opening of the cation channel, and thereby converts the chemical signal to an electrical impulse. The receptor then desensitizes rapidly and enters a transient inactive state, characterized by the presence of bound agonist. In the presence of CACNG4 or CACNG7 or CACNG8, shows resensitization which is characterized by a delayed accumulation of current flux upon continued application of glutamate ().
