Application Notes
PBMC: Add 10 µl of MAB/106 PBMC in 100 µl PBS. Mix gently and incubate for 15 minutes at 2° to 8° C. Wash twice with PBS and analyze or fix with 0.5% v/v of paraformaldehyde in PBS and analyze. WHOLE BLOOD: Add 10 µl of MAB/100 µl of whole blood. Mix gently and incubate for 15 minutes at room temperature (20° C). Lyse the whole blood. Wash once with PBS and analyze or fix with 0.5% v/v of paraformaldehyde in PBS and analyze. See instrument manufacturers instructions for Lysed Whole Blood and Immunofluorescence analysis with a flow cytometer or microscope. ALLOPHYCOCYANIN: (APC) conjugates are analyzed in multi-color flow cytometry with instruments equipped with a second laser and proper filters. Laser excitation is at 633 nm with a Helium Neon (HeNe) laser or a 600-640 nm (633 nm) range for a Dye laser. Peak fluorescence emission is at 660 nm.

Provided as a solution in phosphate buffered saline with 0.08% sodium azide.
Preparation and Storage
Product should be stored at -20 degree C. Aliquot to avoid freeze/thaw cycles. Products are stable for one year from purchase when stored properly.

The CD103 antigen is a 160-130-105 kDa molecule which is expressed on mucosa-associated T lymphocytes and activated cells and on subset of TGF beta -1 cells. This human intra epithelial lymphocyte marker is the E integrin chain associated with the b7 subunit. CD103 antibody stains a few bone marrow cells and, rarely, peripheral blood lymphocytes. CD103 is expressed by 0.5 to 2 % of resting lymphocytes in Peripheral blood and lymphoid organs.

NCBI Description

Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. This gene encodes an I-domain-containing alpha integrin that undergoes post-translational cleavage in the extracellular domain, yielding disulfide-linked heavy and light chains. In combination with the beta 7 integrin, this protein forms the E-cadherin binding integrin known as the human mucosal lymphocyte-1 antigen. This protein is preferentially expressed in human intestinal intraepithelial lymphocytes (IEL), and in addition to a role in adhesion, it may serve as an accessory molecule for IEL activation. Function: Integrin alpha-E/beta-7 is a receptor for E-cadherin. It mediates adhesion of intra-epithelial T-lymphocytes to epithelial cell monolayers.

Subunit structure: Heterodimer of an alpha and a beta subunit. The alpha subunit is composed of a heavy and a light chains linked by a disulfide bond. Alpha-E associates with beta-7.

Subcellular location: Membrane; Single-pass type I membrane protein.

Tissue specificity: Expressed on a subclass of T-lymphocytes known as intra-epithelial lymphocytes which are located between mucosal epithelial cells.

Induction: Integrin alpha-E/beta-7 is induced by TGFB1. Ref.6

Domain: The integrin I-domain (insert) is a VWFA domain. Integrins with I-domains do not undergo protease cleavage.

Sequence similarities: Belongs to the integrin alpha chain family.Contains 7 FG-GAP repeats.Contains 1 VWFA domain.

Researchers should empirically determine the suitability of the CD103 itgae for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD103, Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening.
