Intra-assay Precision
Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level TNFSF14 were tested 20 times on one plate, respectively. Intra-Assay: CV<10%
Inter-assay Precision
Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level TNFSF14 were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Related Product Information for TNFSF14 elisa kit
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of TNFSF14 in mouse serum, plasma, saliva, tissue homogenates and other biological fluids.

Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to TNFSF14. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to TNFSF14. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain TNFSF14, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of TNFSF14 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

The protein encoded by this gene is a member of the tumor necrosis factor (TNF) ligand family. This protein is a ligand for TNFRSF14, which is a member of the tumor necrosis factor receptor superfamily, and which is also known as a herpesvirus entry mediator (HVEM). This protein may function as a costimulatory factor for the activation of lymphoid cells and as a deterrent to infection by herpesvirus. This protein has been shown to stimulate the proliferation of T cells, and trigger apoptosis of various tumor cells. This protein is also reported to prevent tumor necrosis factor alpha mediated apoptosis in primary hepatocyte. Two alternatively spliced transcript variant encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008]

Uniprot Description

TNFSF14: Cytokine that binds to TNFRSF3/LTBR. Binding to the decoy receptor TNFRSF6B modulates its effects. Activates NFKB, stimulates the proliferation of T-cells, and inhibits growth of the adenocarcinoma HT-29. Acts as a receptor for Herpes simplex virus. Belongs to the tumor necrosis factor family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral; Inhibitor; Cytokine

Chromosomal Location of Human Ortholog: 19p13.3

Cellular Component: extracellular space; cytoplasm; integral to membrane; plasma membrane

Molecular Function: protein binding; caspase inhibitor activity; cytokine activity; tumor necrosis factor receptor binding; receptor binding

Biological Process: T cell proliferation; release of cytoplasmic sequestered NF-kappaB; T cell activation; apoptosis; T cell costimulation; immune response; T cell homeostasis; negative regulation of caspase activity; signal transduction; positive regulation of myoblast differentiation

Product Notes

The Mouse TNFSF14 tnfsf14 (Catalog #AAA2021982) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2021982 ELISA Kit recognizes Mouse TNFSF14. It is sometimes possible for the material contained within the vial of "Tumor Necrosis Factor Ligand Superfamily, Member 14 (TNFSF14), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
