Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Mouse Tubby protein homolog (TUB) ELISA Kit employs a two-site sandwich ELISA to quantitate TUB in samples. An antibody specific for TUB has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TUB present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TUB is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TUB bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: To form microtubules, the dimers of alpha- and beta-tubulin bind to GTP and assemble onto the ends of microtubules while in the GTP-bound state. After being incorporated into the microtubule, the bound molecule of GTP will hydrolyse into GDP. Although both subunits bind GTP, only the beta-subunit has GTPase activity; that is, beta-tubulin can hydrolyse GTP to GDP whereas alpha-tubulin cannot. Whether the beta-tubulin member of the tubulin dimer is bound to GTP or GDP influences the stability of the dimer in the microtubule. Dimers bound to GTP tend to assemble into microtubules, while dimers bound to GDP tend to fall apart; thus, this GTP cycle is essential for the dynamic instability of the microtubule. Class III beta-tubulin- is a microtubule element expressed exclusively in neurons, and is a popular identifier specific for neurons in nervous tissue.

Uniprot Description

tubby: an effector protein for heterotrimeric G protein-coupled receptors. Binds phospholipid and is anchored to the plasma membrane through binding phosphatidylinositol-4,5-bisphosphate. Is released upon activation of phospholipase C. Translocates from the plasma membrane to the nucleus upon activation by the G-alpha(q)/(11) subclass of G proteins that are linked to receptors including the seratonin receptor 5-HT(2C). Can bind DNA in vitro and may contribute to the regulation of transcription in the nucleus. TUB can be phosphorylated by the insulin receptor and may participate in insulin receptor signaling. Does not have a cleavable signal peptide and is secreted by a non-conventional pathway. Highly expressed in the paraventricular nucleus of the hypothalamus and several other brain regions. May play a role in the hypothalamic regulation of body weight. Contributes to stimulation of phagocytosis of apoptotic retinal pigment epithelium (RPE) cells and macrophages. Belongs to the TUB family. Mutations in the tubby promote maturity-onset obesity in mice. Interacts with GNAQ. Interacts with TULP1. Two isoforms of the human protein are produced through alternative splicing.
