Description: This assay employs a two-site sandwich ELISA to quantitate SEPT4 in samples. An antibody specific for SEPT4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SEPT4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SEPT4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SEPT4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Septin-4 is a member of the septin gene family of nucleotide binding proteins, originally described in yeast as cell division cycle regulatory proteins. Septins are highly conserved in yeast, Drosophila, and mouse and appear to regulate cytoskeletal organization. The protein encoded by this gene is thought to be part of a complex involved in cytokinesis. Alternatively spliced variants which encode different protein isoforms have been described; however, not all variants have been fully characterized.PNUTL2 contains 478 amino acids and a GTP-binding domain. Sequence comparisons revealed that PNUTL2 shares 88% identity with mouse h5 and 50 to 60% identity with members of the septin family, which are GTP-binding proteins involved in cytokinesis. Northern blot analysis indicated that the 1.7-kb PNUTL2 mRNA is expressed ubiquitously.
