Intended Uses: This sandwich kit is for the accurate quantitative detection of Mouse Nesfatin1 (also known as SOST) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Mouse SOST antibody. SOST present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse SOST Antibody is added and binds to SOST in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated SOST antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse SOST. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

Sclerostin is a secreted glycoprotein with a C-terminal cysteine knot-like (CTCK) domain and sequence similarity to the DAN (differential screening-selected gene aberrative in neuroblastoma) family of bone morphogenetic protein (BMP) antagonists. Loss-of-function mutations in this gene are associated with an autosomal-recessive disorder, sclerosteosis, which causes progressive bone overgrowth. A deletion downstream of this gene, which causes reduced sclerostin expression, is associated with a milder form of the disorder called van Buchem disease. [provided by RefSeq, Jul 2008]
