Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for PDHX elisa kit
Description: This Mouse Pyruvate dehydrogenase protein X component, mitochondrial (PDHX) ELISA Kit employs a two-site sandwich ELISA to quantitate PDHX in samples. An antibody specific for PDHX has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PDHX present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PDHX is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PDHX bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: The PDHX gene encodes component X of the pyruvate dehydrogenase (PDH) complex. For a detailed description of the pyruvate dehydrogenase complex. The mammalian PDH complex differs from that in E. coli and from the other mammalian alpha-keto acid dehydrogenases by the presence of a 53-kD protein called protein X. Component X binds to the E3 component of the PDH complex. The open reading frame encodes an 501-amino acid protein sequence that is 20% identical with that encoded by the yeast PDX1 gene and 40% identical with that encoded by the lipoacyltransferase component E2 of the PDH (E2), and includes a lipoyl-bearing domain. Sequence comparisons showed that the PDHX protein contains an E3-binding domain.

Uniprot Description

PDHX: a mitochondrial protein required for anchoring dihydrolipoamide dehydrogenase (E3) to the pyruvate dehydrogenase complexes (PDHC) of eukaryotes. This specific binding is essential for a PDHC function. Eukaryotic pyruvate dehydrogenase complexes are organized about a core consisting of the oligomeric dihydrolipoamide acetyl-transferase (E2), around which are arranged multiple copies of pyruvate dehydrogenase (E1), dihydrolipoamide dehydrogenase (E3), and protein X bound by non-covalent bonds. Belongs to the 2-oxoacid dehydrogenase family. Contains 1 lipoyl-binding domain.

Protein type: Mitochondrial

Cellular Component: mitochondrion

Molecular Function: transferase activity, transferring acyl groups

Biological Process: metabolic process

Product Notes

The Mouse PDHX pdhx (Catalog #AAA9718061) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9718061 ELISA Kit recognizes Mouse PDHX. It is sometimes possible for the material contained within the vial of "Pyruvate Dehydrogenase Protein X Component, Mitochondrial (PDHX), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
