Principle of the assay: LHB ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-LHB antibody and an LHB-HRP conjugate. The assay sample and buffer are incubated together with LHB-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the LHB concentration since LHB from samples and LHB-HRP conjugate compete for the anti-LHB antibody binding site. Since the number of sites is limited, as more sites are occupied by LHB from the sample, fewer sites are left to bind LHB-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The LHB concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is a member of the glycoprotein hormone beta chain family and encodes the beta subunit of luteinizing hormone (LH). Glycoprotein hormones are heterodimers consisting of a common alpha subunit and an unique beta subunit which confers biological specificity. LH is expressed in the pituitary gland and promotes spermatogenesis and ovulation by stimulating the testes and ovaries to synthesize steroids. The genes for the beta chains of chorionic gonadotropin and for luteinizing hormone are contiguous on chromosome 19q13.3. Mutations in this gene are associated with hypogonadism which is characterized by infertility and pseudohermaphroditism. [provided by RefSeq, Jul 2008]
