Principle of the Assay: The Picokine™ Mouse KLKB1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Mouse KLKB1 with a 96-well strip plate that is pre-coated with antibody specific for KLKB1. The detection antibody is a biotinylated antibody specific for KLKB1. The capture antibody is a monoclonal antibody from rat, the detection antibody is a biotinylated monoclonal antibody from goat. The kit contains recombinant Mouse KLKB1 with immunogen: Expression system for standard: NSO, Immunogen sequence: G20-A638. The kit is analytically validated with ready to use reagents. To measure Mouse KLKB1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Mouse KLKB1 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse KLKB1 in the sample.!!Background/Introduction: Plasma kallikrein is a protein that in humans is encoded by the KLKB1 gene. It is mapped to 4q35.2. This gene encodes a glycoprotein that participates in the surface-dependent activation of blood coagulation, fibrinolysis, kinin generation and inflammation. The encoded preproprotein present in plasma as a non-covalent complex with high molecular weight kininogen undergoes proteolytic processing mediated by activated coagulation factor XII to generate a disulfide-linked, heterodimeric serine protease comprised of heavy and light chains. Certain mutations in this gene cause prekallikrein deficiency. Alternative splicing results in multiple transcript variants encoding different isoforms.

KLKB1: The enzyme cleaves Lys-Arg and Arg-Ser bonds. It activates, in a reciprocal reaction, factor XII after its binding to a negatively charged surface. It also releases bradykinin from HMW kininogen and may also play a role in the renin-angiotensin system by converting prorenin into renin. Defects in KLKB1 are the cause of prekallikrein deficiency (PKK deficiency); also known as Fletcher factor deficiency. This disorder is a blood coagulation defect. Belongs to the peptidase S1 family. Plasma kallikrein subfamily.
