Intended Uses: This sandwich kit is for the accurate quantitative detection of Mouse Insulin (also known as INS) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.
Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Mouse INS antibody. INS present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse INS Antibody is added and binds to INS in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated INS antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse INS. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

After removal of the precursor signal peptide, proinsulin is post-translationally cleaved into three peptides: the B chain and A chain peptides, which are covalently linked via two disulfide bonds to form insulin, and C-peptide. Binding of insulin to the insulin receptor (INSR) stimulates glucose uptake. A multitude of mutant alleles with phenotypic effects have been identified. There is a read-through gene, INS-IGF2, which overlaps with this gene at the 5' region and with the IGF2 gene at the 3' region. Alternative splicing results in multiple transcript variants.
