Mouse Histone-lysine N-methyltransferase EHMT1 (EHMT1) ELISA Kit
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate EHMT1 in Mouse serum, plasma, tissue homogenates. An antibody specific for EHMT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any EHMT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EHMT1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EHMT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background: EHMT1 is a histone methyltransferase that is part of the E2F6 complex, which represses transcription. The encoded protein methylates the Lys-9 position of histone H3, which tags it for transcriptional repression. This protein may be involved in the silencing of MYC- and E2F-responsive genes and therefore could play a role in the G0/G1 cell cycle transition. The 1,267-amino acid EU-HMTase-1 protein contains ankyrin repeats and a SET domain. EU-HMTase-1 shares 63% sequence similarity with NG36/G9A. EU-HMTase-1 exhibited histone methyltransferase activity with a specificity for histone H3 in core histones, whereas it hardly methylated nucleosomes, suggesting that other subunits in the E2F6 complex are required for modification of nucleosomal substrates.
NCBI and Uniprot Product Information
Uniprot Description
Histone methyltransferase that specifically mono- and dimethylates 'Lys-9' of histone H3 (H3K9me1 and H3K9me2, respectively) in euchromatin. H3K9me represents a specific tag for epigenetic transcriptional repression by recruiting HP1 proteins to methylated histones. Also weakly methylates 'Lys-27' of histone H3 (H3K27me). Also required for DNA methylation, the histone methyltransferase activity is not required for DNA methylation, suggesting that these 2 activities function independently. Probably targeted to histone H3 by different DNA-binding proteins like E2F6, MGA, MAX and/or DP1. During G0 phase, it probably contributes to silencing of MYC- and E2F-responsive genes, suggesting a role in G0/G1 transition in cell cycle. In addition to the histone methyltransferase activity, also methylates non-histone proteins: mediates dimethylation of 'Lys-373' of p53/TP53.

Product Notes
The Mouse EHMT1 ehmt1 (Catalog #AAA287696) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA287696 ELISA Kit recognizes Mouse EHMT1. It is sometimes possible for the material contained within the vial of "Histone-lysine N-methyltransferase EHMT1 (EHMT1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
