Description: This assay employs a two-site sandwich ELISA to quantitate GTDC1 in samples. An antibody specific for GTDC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any GTDC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GTDC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GTDC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Zhao et al. (2004) cloned GTDC1 from a fetal brain cDNA library as part of a high throughput DNA sequencing analysis. The deduced 458-amino acid GTDC1 protein has a calculated molecular mass of 52.6 kD and shares 75% and 37% identity with the putative murine and Drosophila homologs, respectively. GTDC1 contains an N-terminal transmembrane region, which predicts a type II transmembrane protein, and a C-terminal glycosyltransferase-like domain. RT-PCR analysis detected ubiquitous expression of GTDC1 mRNA, at relatively high levels in lung, spleen, testis, and peripheral blood leukocytes.
