Principle of the Assay: RAD18 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-RAD18 antibody and an RAD18-HRP conjugate. The assay sample and buffer are incubated together with RAD18-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RAD18 concentration since RAD18 from samples and RAD18-HRP conjugate compete for the anti-RAD18 antibody binding site. Since the number of sites is limited, as more sites are occupied by RAD18 from the sample, fewer sites are left to bind RAD18-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RAD18 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is highly similar to S. cerevisiae DNA damage repair protein Rad18. Yeast Rad18 functions through its interaction with Rad6, which is an ubiquitin-conjugating enzyme required for post-replication repair of damaged DNA. Similar to its yeast counterpart, this protein is able to interact with the human homolog of yeast Rad6 protein through a conserved ring-finger motif. Mutation of this motif results in defective replication of UV-damaged DNA and hypersensitivity to multiple mutagens. [provided by RefSeq, Jul 2008]
