Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Mouse DEFB 104A antibody. DEFB 104A present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse DEFB 104A Antibody is added and binds to DEFB 104A in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated DEFB 104A antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse DEFB 104A. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Defensins form a family of antimicrobial and cytotoxic peptides made by neutrophils. Defensins are short, processed peptide molecules that are classified by structure into three groups: alpha-defensins, beta-defensins and theta-defensins. All beta-defensin genes are densely clustered in four to five syntenic chromosomal regions. Chromosome 8p23 contains at least two copies of the duplicated beta-defensin cluster. This duplication results in two identical copies of defensin, beta 104, DEFB104A and DEFB104B, in head-to-head orientation. This gene, DEFB104A, represents the more centromeric copy.
