Intended Uses: This CNGB3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse CNGB3. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CNGB3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CNGB3 antibody and an CNGB3-HRP conjugate. The assay sample and buffer are incubated together with CNGB3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CNGB3 concentration since CNGB3 from samples and CNGB3-HRP conjugate compete for the anti-CNGB3 antibody binding site. Since the number of sites is limited, as more sites are occupied by CNGB3 from the sample, fewer sites are left to bind CNGB3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CNGB3 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes the beta subunit of a cyclic nucleotide-gated ion channel. The encoded beta subunit appears to play a role in modulation of channel function in cone photoreceptors. This heterotetrameric channel is necessary for sensory transduction, and mutations in this gene have been associated with achromatopsia 3, progressive cone dystrophy, and juvenile macular degeneration, also known as Stargardt Disease. [provided by RefSeq, Feb 2010]

Uniprot Description

CNGB3: Visual signal transduction is mediated by a G-protein coupled cascade using cGMP as second messenger. This protein can be activated by cGMP which leads to an opening of the cation channel and thereby causing a depolarization of rod photoreceptors. Induced a flickering channel gating, weakened the outward rectification in the presence of extracellular calcium, increased sensitivity for L-cis diltiazem and enhanced the cAMP efficiency of the channel when coexpressed with CNGA3. Essential for the generation of light-evoked electrical responses in the red-, green- and blue sensitive cones. Defects in CNGB3 are the cause of Stargardt disease type 1 (STGD1). STGD is one of the most frequent causes of macular degeneration in childhood. It is characterized by macular dystrophy with juvenile-onset, rapidly progressive course, alterations of the peripheral retina, and subretinal deposition of lipofuscin-like material. STGD1 inheritance is autosomal recessive. Defects in CNGB3 are the cause of achromatopsia type 3 (ACHM3); also known as Pingelapese blindness. ACHM3 is a congenital complete achromatopsia and is distinct from total colorblindness mainly because of the consistent concurrence of severe myopia. Belongs to the cyclic nucleotide-gated cation channel (TC 1.A.1.5) family. CNGB3 subfamily. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Mouse CNGB3 cngb3 (Catalog #AAA7222591) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7222591 ELISA Kit recognizes Mouse CNGB3. It is sometimes possible for the material contained within the vial of "Cyclic nucleotide-gated cation channel beta-3 (CNGB3), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
