Intended Uses: This CRY2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse CRY2. This ELISA kit for research use only!

Principle of the Assay: CRY2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CRY2 antibody and an CRY2-HRP conjugate. The assay sample and buffer are incubated together with CRY2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CRY2 concentration since CRY2 from samples and CRY2-HRP conjugate compete for the anti-CRY2 antibody binding site. Since the number of sites is limited, as more sites are occupied by CRY2 from the sample, fewer sites are left to bind CRY2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CRY2 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a flavin adenine dinucleotide-binding protein that is a key component of the circadian core oscillator complex, which regulates the circadian clock. This gene is upregulated by CLOCK/ARNTL heterodimers but then represses this upregulation in a feedback loop using PER/CRY heterodimers to interact with CLOCK/ARNTL. Polymorphisms in this gene have been associated with altered sleep patterns. The encoded protein is widely conserved across plants and animals. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Feb 2014]

Uniprot Description

CRY2: Blue light-dependent regulator of the circadian feedback loop. Inhibits CLOCK|NPAS2-ARNTL E box-mediated transcription. Acts, in conjunction with CRY2, in maintaining period length and circadian rhythmicity. Has no photolyase activity. Capable of translocating circadian clock core proteins such as PER proteins to the nucleus. May inhibit CLOCK|NPAS2-ARNTL transcriptional activity through stabilizing the unphosphorylated form of ARNTL. Belongs to the DNA photolyase class-1 family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Lyase; DNA-binding

Chromosomal Location of Human Ortholog: 11p11.2

Cellular Component: cytoplasm; extracellular region; nucleus

Molecular Function: ubiquitin binding; protein binding; DNA binding; DNA (6-4) photolyase activity; damaged DNA binding; deoxyribodipyrimidine photo-lyase activity; blue light photoreceptor activity; single-stranded DNA binding; phosphatase binding

Biological Process: circadian rhythm; photoreactive repair; transcription, DNA-dependent; negative regulation of transcription from RNA polymerase II promoter; regulation of circadian rhythm; glucose homeostasis; protein-chromophore linkage; entrainment of circadian clock by photoperiod; blue light signaling pathway; negative regulation of phosphoprotein phosphatase activity; negative regulation of circadian rhythm; circadian regulation of gene expression; negative regulation of transcription, DNA-dependent

Product Notes

The Mouse CRY2 cry2 (Catalog #AAA7248707) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7248707 ELISA Kit recognizes Mouse CRY2. It is sometimes possible for the material contained within the vial of "Cryptochrome-2 (CRY2), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
