Principle of the assay: CLEC12A ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CLEC12A antibody and an CLEC12A-HRP conjugate. The assay sample and buffer are incubated together with CLEC12A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLEC12A concentration since CLEC12A from samples and CLEC12A-HRP conjugate compete for the anti-CLEC12A antibody binding site. Since the number of sites is limited, as more sites are occupied by CLEC12A from the sample, fewer sites are left to bind CLEC12A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLEC12A concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily. Members of this family share a common protein fold and have diverse functions, such as cell adhesion, cell-cell signaling, glycoprotein turnover, and roles in inflammation and immune response. The protein encoded by this gene is a negative regulator of granulocyte and monocyte function. Several alternatively spliced transcript variants of this gene have been described, but the full-length nature of some of these variants has not been determined. This gene is closely linked to other CTL/CTLD superfamily members in the natural killer gene complex region on chromosome 12p13. [provided by RefSeq, May 2011]
