Intended Uses: This ATHL1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ATHL1. This ELISA kit for research use only!

Principle of the Assay: ATHL1 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-ATHL1 antibody and an ATHL1-HRP conjugate. The assay sample and buffer are incubated together with ATHL1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATHL1 concentration since ATHL1 from samples and ATHL1-HRP conjugate compete for the anti-ATHL1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ATHL1 from the sample, fewer sites are left to bind ATHL1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATHL1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a bifunctional protein that catalyzes the last two steps of the de novo purine biosynthetic pathway. The N-terminal domain has phosphoribosylaminoimidazolecarboxamide formyltransferase activity, and the C-terminal domain has IMP cyclohydrolase activity. A mutation in this gene results in AICA-ribosiduria. [provided by RefSeq, Sep 2009]

A neurologically devastating inborn error of purine biosynthesis. Patients excrete massive amounts of AICA-riboside in the urine and accumulate AICA- ribotide and its derivatives in erythrocytes and fibroblasts. AICAR causes profound mental retardation, epilepsy, dysmorphic features and congenital blindness. Belongs to the PurH family.
