Specificity
This assay has high sensitivity and excellent specificity for detection of AGPAT3. No significant cross-reactivity or interference between AGPAT3 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between AGPAT3 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This AGPAT3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of MouseAGPAT3. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: AGPAT3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-AGPAT3 antibody and an AGPAT3-HRP conjugate. The assay sample and buffer are incubated together with AGPAT3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AGPAT3 concentration since AGPAT3 from samples and AGPAT3-HRP conjugate compete for the anti-AGPAT3 antibody binding site. Since the number of sites is limited, as more sites are occupied by AGPAT3 from the sample, fewer sites are left to bind AGPAT3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AGPAT3 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is an acyltransferase that converts lysophosphatidic acid into phosphatidic acid, which is the second step in the de novo phospholipid biosynthetic pathway. The encoded protein may be an integral membrane protein. Two transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Jul 2008]

Uniprot Description

AGPAT3: Converts lysophosphatidic acid (LPA) into phosphatidic acid by incorporating an acyl moiety at the sn-2 position of the glycerol backbone. Belongs to the 1-acyl-sn-glycerol-3-phosphate acyltransferase family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Mouse AGPAT3 agpat3 (Catalog #AAA7200885) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7200885 ELISA Kit recognizes Mouse AGPAT3. It is sometimes possible for the material contained within the vial of "1-acyl-sn-glycerol-3-phosphate acyltransferase gamma (AGPAT3), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
