Application Notes
TL2.1 antibody has been reported as an effective inhibitor of TLR2-dependent cytokine production and might be used to control and manipulate signalling pathways related to TLR2 (Flo et al., 2000). This antibody has also been exstensively used in flow cytometry and TL2.1 is recommended for the use in flow cytometric investigation of TLR2 expression (Jamin et al., 2015; Wang et al., 2016). For instance, one group treated dendritic cells with azithromycin and observed that this substance supressed TLR2 expression detected via flow cytometry (Lin et al., 2016). Finally, this antibody has been successfully applied in immunofluorescence analysis of TLR2 expression in various cells. One group utilised TL2.1 antibody to investigate TLR2 expression (among others pathogen-related receptors) on SRIK-NKL cells. CD8+ NK/T cell line (Srivastava and Srivastava, 2005).

NCBI Description

The protein encoded by this gene is a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This gene is expressed most abundantly in peripheral blood leukocytes, and mediates host response to Gram-positive bacteria and yeast via stimulation of NF-kappaB. [provided by RefSeq, Jul 2008]

TLR2: Cooperates with LY96 to mediate the innate immune response to bacterial lipoproteins and other microbial cell wall components. Cooperates with TLR1 to mediate the innate immune response to bacterial lipoproteins or lipopeptides. Acts via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. May also promote apoptosis in response to lipoproteins. Recognizes mycoplasmal macrophage- activating lipopeptide-2kD (MALP-2), soluble tuberculosis factor (STF), phenol-soluble modulin (PSM) and B.burgdorferi outer surface protein A lipoprotein (OspA-L) cooperatively with TLR6. Researchers should empirically determine the suitability of the TLR2 tlr2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
