Preparation and Storage
This product is shipped at ambient temperature. It is recommended to aliquot and store at -20 degree C on receipt. When thawed, aliquot the sample as needed. Keep aliquots at 2-8 degree C for short term use (up to 4 weeks) and store the remaining aliquots at -20 degree C.

Avoid repeated freezing and thawing as this may denature the antibody. Storage in frost-free freezers is not recommended.

Mouse anti Human TIMP-1 antibody, clone M05, recognizes Tissue Inhibitor of Metalloproteinases 1 (TIMP-1) also known as metalloproteinase inhibitor 1, EPA and fibroblast collagenase inhibitor.

TIMP-1 is a member of the TIMP family of proteins that assist in regulating the activity of matrix metalloproteinases (MMPs) and it is mainly expressed by macrophages and fibroblasts. TIMP proteins are specific endogenous inhibitors that bind to the active site of MMPs in a stoichiometric 1:1 molar ratio, thereby blocking access to ECM substrates (Kapoor et al. 2016, Robert et al. 2016). The TIMP proteins including TIMP-1 have poor specificity for any particular MMP and each can inhibit multiple MMPs with differing efficacies (Cui et al. 2017).

The ratio of MMP and TIMP often determine the extent of ECM protein degradation and tissue remodeling. An imbalance of this ratio such as with decreased expression of TIMP can result in various pathological conditions, most notably, heart failure, osteoarthritis and cancer. Low levels of TIMP have also been associated with slow-to-heal wounds and venous wounds (Cui et al. 2017).

The biotinylated Mouse anti Human TIMP-1 antibody, clone M05 can be used as a detection antibody in a sandwich ELISA with the purified Mouse anti Human TIMP-1 antibody, clone M03 as the capture antibody.

NCBI Description

This gene belongs to the TIMP gene family. The proteins encoded by this gene family are natural inhibitors of the matrix metalloproteinases (MMPs), a group of peptidases involved in degradation of the extracellular matrix. In addition to its inhibitory role against most of the known MMPs, the encoded protein is able to promote cell proliferation in a wide range of cell types, and may also have an anti-apoptotic function. Transcription of this gene is highly inducible in response to many cytokines and hormones. In addition, the expression from some but not all inactive X chromosomes suggests that this gene inactivation is polymorphic in human females. This gene is located within intron 6 of the synapsin I gene and is transcribed in the opposite direction. [provided by RefSeq, Jul 2008]

TIMP1: Complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them by binding to their catalytic zinc cofactor. Also mediates erythropoiesis in vitro; but, unlike IL-3, it is species-specific, stimulating the growth and differentiation of only human and murine erythroid progenitors. Known to act on MMP-1, MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-10, MMP-11, MMP-12, MMP-13 and MMP-16. Does not act on MMP-14. Belongs to the protease inhibitor I35 (TIMP) family.
