Immunoprecipitation (IP)

(TIM50 was immunoprecipitated from 1mg of Jurkat cells membrane fraction, blotted with M25108 of 10 ug. Western blot was performed from the immunopre-cipitate using M25108 at 1/1000 dilution. Anti-Mouse-IgG(HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/10000 dilution. Lane 1: Jurkat cells membrane fraction. Lane2: IP product of Jurkat cells membrane fraction.Predicted band size : 40 KDaObserved band size : 40 KDaBlocking and dilution buffer and concentration:5% milk/TBST.)

Flow Cytometry (FC/FACS)

(Flow cytometric analysis of 2% paraformaldehyde-fixed Jurkat (Human T cell leukemia cells from peripheral blood) cells labeling TIM50 with M25108 at 1/200 dilution (red) compared with a mouse monoclonal IgG isotype control (black) and an unlabelled control (cells without incubation with primary antibody, blue). Goat anti-mouse IgG (FITC) at 1/300 dilution was used as the secondary antibody.)

NCBI Description

This gene encodes a subunit of the TIM23 inner mitochondrial membrane translocase complex. The encoded protein functions as the receptor subunit that recognizes the mitochondrial targeting signal, or presequence, on protein cargo that is destined for the mitochondrial inner membrane and matrix. This protein may also play a role in maintaining the membrane permeability barrier, and knockdown of this gene in human cells results in the release of cytochrome c and apoptosis.
