Application Notes
Western Blot: This antibody can be used at 1-2ug/ml with the appropriate secondary reagents to detect human TEM8. Using a colorimetric detection system, the detection limit for rhTEM8 is approximately 5 ng/lane under non-reducing and reducing conditions. Chemiluminescent detection will increase sensitivity by 5 to 50 fold., Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect human TEM8. The detection limit for rhTEM8 is approximately 5 ng/well., Optimal dilutions should be determined by each laboratory for each application.

Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Tumor Endothelial Marker 8 (TEM8), also called ATR1, is a glycoprotein of the anthrax toxin receptor family that is expressed by endothelial cells. Type I transmembrane isoforms of 564 aa (80-85 kD) and 368 aa (60 kD) and potentially secreted isoforms of 330 aa and 297 aa (45 kD) are differentially expressed. All diverge at the C-terminal end but share the N-terminal vWFA domain that binds the anthrax toxin PA subunit as well as collagens I and IV. Human TEM8 shares 99% aa identity with mouse and rat and 92% identity with dog and chick TEM8 within the extracellular domain.

NCBI Description

This gene encodes a type I transmembrane protein and is a tumor-specific endothelial marker that has been implicated in colorectal cancer. The encoded protein has been shown to also be a docking protein or receptor for Bacillus anthracis toxin, the causative agent of the disease, anthrax. The binding of the protective antigen (PA) component, of the tripartite anthrax toxin, to this receptor protein mediates delivery of toxin components to the cytosol of cells. Once inside the cell, the other two components of anthrax toxin, edema factor (EF) and lethal factor (LF) disrupt normal cellular processes. Three alternatively spliced variants that encode different protein isoforms have been described. [provided by RefSeq]

ANTXR1: Plays a role in cell attachment and migration. Interacts with extracellular matrix proteins and with the actin cytoskeleton. Mediates adhesion of cells to type 1 collagen and gelatin, reorganization of the actin cytoskeleton and promotes cell spreading. Plays a role in the angiogenic response of cultured umbilical vein endothelial cells. Defects in ANTXR1 are associated with susceptibility to hemangioma capillary infantile (HCI). HCI are benign, highly proliferative lesions involving aberrant localized growth of capillary endothelium. They are the most common tumor of infancy, occurring in up to 10% of all births. Hemangiomas tend to appear shortly after birth and show rapid neonatal growth for up to 12 months characterized by endothelial hypercellularity and increased numbers of mast cells. This phase is followed by slow involution at a rate of about 10% per year and replacement by fibrofatty stroma. Belongs to the ATR family. 4 isoforms of the human protein are produced by alternative splicing.
