Chromatin Immunoprecipitation (ChIP) (Suz12 antibody (mAb) tested by ChIP. Chromatin IP performed using the ChIP-IT Express Kit and HeLa chromatin (1.5 x 106 cell equivalents per ChIP) using 10 ug of Suz12 antibody or the equivalent amount of rabbit IgG as a negative control. Real time, quantitative PCR (RT-qPCR) was performed on DNA purified from each of the ChIP reactions using a primer pair specific for the Myt1 gene promoter. Data are presented as Fold Enrichment of the ChIP antibody signal versus the negative control IgG using the ddCT method.)

Short Description: Our Suz12 antibody (mAb)(Clone 2AO9) was raised in a Mouse host. It has been validated for use in Chromatin Immunoprecipitation, Immunohistochemistry and Immunoprecipitation; it has been shown to react with Human samples.

Background: Suz12 (Suppressor-of-Zeste 12 protein, also designated as ChET9, JJAZ1 protein) is a component of the Polycomb group complexes 2, 3 and 4 (PRC2, PRC3 and PRC4), which also contain the histone methyltransferase EZH2, EED (Embryonic Ectoderm Development protein) protein and the histone-binding proteins RbAP46 and RbAP48. PRC2 is responsible for di- and trimethylation of histone 3 at lysine 27 (Histone H3K27me2 and Histone H3K27me3), which leads to transcriptional repression of the target genes. Suz12 is a nuclear protein belonging to the VEFS family. Suz12 contains a zinc-finger binding motif and a VEFS box, which is required for the interaction between Suz12 and EZH2.

NCBI Description

This zinc finger gene has been identified at the breakpoints of a recurrent chromosomal translocation reported in endometrial stromal sarcoma. Recombination of these breakpoints results in the fusion of this gene and JAZF1. The protein encoded by this gene contains a zinc finger domain in the C terminus of the coding region. [provided by RefSeq, Jul 2009]

Product Notes

The Suz12 suz12 (Catalog #AAA388616) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Suz12 antibody (mAb)(Clone 2AO9) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Suz12 can be used in a range of immunoassay formats including, but not limited to, Chromatin Immunoprecipitation (ChIP), Immunohistochemistry (IHC), Immunoprecipitation (IP). Applications Validated: ChIP: 2.5-10ug per ChIP This antibody has been published, but has not been validated, for Western blot. For an antibody validated for use in Western blotting. Researchers should empirically determine the suitability of the Suz12 suz12 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Suz12, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
