Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20 degree C or colder. Aliquots are stable for at least 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

SLPI is a 11.7kD cationic inhibitor of neutrophil elastase and to a lesser extent of cathepsin G. It is locally produced by epithelial cells in the lung, skin and other organs and by Polymorphonuclear leukocytes (PMN) and (in mice) by macrophages. In addition to its proteinase inhibitory properties that may serve to protect against proteolytic injury, it was recently shown that SLPI also displays several other functions such as antimicrobial and anti-inflammatory activities. These appear to be independent of its ability to inhibit PMN serine proteinases. SLPI has also been demonstrated to display antibacterial and antifungal activity at concentrations in which SLPI is present in mucosal secretions including those of the lung. Another possible role for SLPI is inhibition of protein-disulfide isomerase that is considered essential for invasion of a cell by the Human Immunodeficiency Virus (HIV).

NCBI Description

This gene encodes a secreted inhibitor which protects epithelial tissues from serine proteases. It is found in various secretions including seminal plasma, cervical mucus, and bronchial secretions, and has affinity for trypsin, leukocyte elastase, and cathepsin G. Its inhibitory effect contributes to the immune response by protecting epithelial surfaces from attack by endogenous proteolytic enzymes; the protein is also thought to have broad-spectrum anti-biotic activity.
