NCBI Description

The incorporation of selenocysteine into a protein requires the concerted action of an mRNA element called a sec insertion sequence (SECIS), a selenocysteine-specific translation elongation factor and a SECIS binding protein. With these elements in place, a UGA codon can be decoded as selenocysteine. The gene described in this record encodes a nuclear protein that functions as a SECIS binding protein. Mutations in this gene have been associated with a reduction in activity of a specific thyroxine deiodinase, a selenocysteine-containing enzyme, and abnormal thyroid hormone metabolism. [provided by RefSeq]

Uniprot Description

SECISBP2: Binds to the SECIS element in the 3'-UTR of some mRNAs encoding selenoproteins. Binding is stimulated by SELB. Defects in SECISBP2 are a cause of abnormal thyroid hormone metabolism (ATHYHM). This phenotype is associated with a reduction in type II iodothyronine deiodinase activity. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Translation

Chromosomal Location of Human Ortholog: 9q22.2

Cellular Component: nucleoplasm; mitochondrion; ribonucleoprotein complex

Molecular Function: ribonucleoprotein binding; mRNA 3'-UTR binding

Biological Process: selenocysteine incorporation

Disease: Thyroid Hormone Metabolism, Abnormal

Research Articles on SECISBP2

1. Observational study of gene-disease association. (HuGE Navigator)
2. Results demonstrate that SECIS-binding protein 2 is required for protection against reactive oxygen species-induced cellular damage and cell survival.
3. SBP2 gene mutation producing early arrest in synthesis of full-length molecule. Demonstration that SBP2 isoforms containing all functional domains could be synthesized from three downstream ATGs explains the relatively mild phenotype caused by this defect
4. ca. 70% of the SBP2 sequence is disordered, whereas the RNA binding domain appears to be folded and functional.
5. Seleniuim is obviously not a limiting factor in the SBP2 deficient individuals when regular daily selenium intake is provided
6. CUG-BP1 and HuR act as factors that bind to the SBP2 3' UTR, which suggests that TTR-RBPs play a role in the regulation of SBP2
7. SECIS binding induces a conformational change in SBP2 that recruits eEFSec, which in concert with the Sec incorporation domain gains access to the ribosomal A site
8. A report of a complex splicing pattern in the 5'-region of human SECISBP2, wherein at least eight splice variants encode five isoforms with varying N-terminal sequence.
9. Data suggest that SBP2 is a major determinant in dictating the hierarchy of selenoprotein synthesis via differential selenoprotein mRNA translation and sensitivity to nonsense-mediated decay.
10. Oxidative stress induces nuclear accumulation of SBP2 via oxidation of cysteine residues within a redox-sensitive cysteine-rich domain.

Product Notes

The SECISBP2 secisbp2 (Catalog #AAA606332) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The SECIS Binding Protein 2 (SECISBP2, DKFZp686C09169, SBP2) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's SECIS Binding Protein 2 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Gel Shift Assay (GS/EMSA). Suitable for use in Dot Blot and Western Blot. Researchers should empirically determine the suitability of the SECISBP2 secisbp2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SECIS Binding Protein 2, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
