Proteins in this family participate in pathways regulated by reversible phosphorylation at serine and threonine residues, potentially playing a role in the regulation of cell growth and differentiation. PP5 or PPT is predominantly a nuclear protein and interacts with CDC16 and CDC27. It is found in association with several proteins that influence intracellular signlaing-cascades intitiated by hormones (glucocorticoids) or cellular stress (hypoxia, oxidative stress and DNA Damage). It phosphorylates serine residues of skeletal muscle phosphorylase and histone H1. It may also be involved in mitosis and RNA biogenesis regulation (1,2).

NCBI Description

This gene encodes a serine/threonine phosphatase which is a member of the protein phosphatase catalytic subunit family. Proteins in this family participate in pathways regulated by reversible phosphorylation at serine and threonine residues; many of these pathways are involved in the regulation of cell growth and differentiation. The product of this gene has been shown to participate in signaling pathways in response to hormones or cellular stress, and elevated levels of this protein may be associated with breast cancer development. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Feb 2011]

PPP5C: a Ser/Thr phosphatase. Plays a role in the survival of cells in a low oxygen stress by suppressing a hypoxia-induced ASK-1/MKK4/JNK signaling cascade that promotes apoptosis. Has been reported to interact with DNA-PK, dephosphorylating it at two functional sites. May play a role in the regulation of RNA biogenesis and/or mitosis.
