Background: Perforin is a 70kDa protein containing 534 amino acid.

Perforin is involved in the killing function by CTLs and NKs. Upon degranulation, perforin inserts itself into the target cell's plasma membrane, forming a pore. It leads to osmotic lysis of the target cells and subsequently enables granzymes to enter the target cells and activate apoptosis, the cell death program.

Perforin has structural and functional similarities to complement component 6, 7, 8 and 9 (C6, C7, C8, C9). Like C9, this protein creates transmembrane tubules and is capable of lysing non-specifically a variety of target cells. This protein is one of the main cytolytic proteins of cytolytic granules, and it is known to be a key effector molecule for T-cell- and natural killer-cell-mediated cytolysis.

Its role in the immune response against tumors and virus infection has been demonstrated.

NCBI Description

The protein encoded by this gene has structural and functional similarities to complement component 9 (C9). Defects in this gene cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare and lethal autosomal recessive disorder of early childhood. Alternative splicing results in multiple transcript variants encoding the same protein. [provided by RefSeq, Jul 2008]

Uniprot Description

PRF1: Plays a key role in secretory granule-dependent cell death, and in defense against virus-infected or neoplastic cells. Plays an important role in killing other cells that are recognized as non-self by the immune system, e.g. in transplant rejection or some forms of autoimmune disease. Can insert into the membrane of target cells in its calcium-bound form, oligomerize and form large pores. Promotes cytolysis and apoptosis of target cells by facilitating the uptake of cytotoxic granzymes. Monomer, as sobluble protein. Homooligomer. Oligomerization is required for pore formation. Repressed by contact with target cells. Belongs to the complement C6/C7/C8/C9 family.

Product Notes

The PRF1 prf1 (Catalog #AAA335420) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Mouse Anti-Human Perforin Monoclonal Antibody, PerCP Conjugated Clone B-D48 reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Perforin can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC). Working Dilution: Use 10 ul to label 0,5.106 cells or 50 ul of whole blood Sodium azide is a poisenous and hazardous substance, handle and dispose of properly. Researchers should empirically determine the suitability of the PRF1 prf1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Perforin, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
