Flow Cytometry (FC/FACS)

(Fig-2: Intracellular flow analysis of Pax8 in Raji cells using 0.5 ug/10^6 cells of antibody (Clone: ABM5F33). Green represents isotype control; red represents anti-Pax8 antibody. Goat anti-mouse PE conjugate was used as secondary antibody. (Cells were fixed with 4% paraformaldehyde for 10 min and washed with PBS by centrifuging at 1100 for 5 min followed by permeabilization for 20 min and washed again as mentioned above. Then cell were incubated with primary antibody for 45 min. and after washing the cells twice in PBS, incubated with conjugated secondary antibody for 30 min. Data acquisition was done after washing twice with PBS as mentioned above).)

NCBI Description

This gene encodes a member of the paired box (PAX) family of transcription factors. Members of this gene family typically encode proteins that contain a paired box domain, an octapeptide, and a paired-type homeodomain. This nuclear protein is involved in thyroid follicular cell development and expression of thyroid-specific genes. Mutations in this gene have been associated with thyroid dysgenesis, thyroid follicular carcinomas and atypical follicular thyroid adenomas. Alternatively spliced transcript variants encoding different isoforms have been described. [provided by RefSeq, Mar 2010]

PAX8: Transcription factor for the thyroid-specific expression of the genes exclusively expressed in the thyroid cell type, maintaining the functional differentiation of such cells. Interacts with WWTR1. Expressed in the excretory system, thyroid gland and Wilms tumors. 5 isoforms of the human protein are produced by alternative splicing.
