Application Notes
Suitable for use in Flow Cytometry, ELISA and Western Blot.
Dilution: Western Blot: 1-2ug/ml. The detection limit is ~10ng/lane under non-reducing conditions. Reducing conditions not recommended.
Flow Cytometry: 25ug/ml. 10ul labels 1-2.5 x 10e5 cells in a total reaction volume 200ul. The binding of unlabeled monoclonal antibodies may be visualized by adding 10ul of a 25ug/ml stock solution of a secondary developing reagent such as goat anti-rat IgG conjugated to a fluorochrome.
Direct ELISA: 0.5-1ug/ml. The detection limit is ~5ng/well.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Mouse PIR-B is a 125kD type I transmembrane glycoprotein with six Ig-like domains in its extracellular domain (ECD) and four ITIM-like sequences in its cytoplasmic domain. The ECD of PIR-B is highly homologous to the ECDs of multiple mouse PIR-A receptors (92-99% aa sequence homology), which have short cytoplasmic tails lacking ITIM motifs. PIR-A receptors have a charged residue in their transmembrane domain that facilitates interaction with ITAM-containing adaptor molecules. Whereas PIR-A receptors deliver activation signals, PIR-B can inhibit receptor-mediated activation signaling. PIR-A and PIR-B have been shown to bind various mouse MHC class I molecules. They have been proposed to be orthologs of human leukocyte immunoglobulin-like receptors.

NCBI Description

Cell signaling pathways rely on a dynamic interaction between activating and inhibiting processes. SHP-1-mediated dephosphorylation of protein tyrosine residues is central to the regulation of several cell signaling pathways. Two types of inhibitory receptor superfamily members are immunoreceptor tyrosine-based inhibitory motif (ITIM)-bearing receptors and their non-ITIM-bearing, activating counterparts. Control of cell signaling via SHP-1 is thought to occur through a balance between PILRalpha-mediated inhibition and PILRbeta-mediated activation. These paired immunoglobulin-like receptor genes are located in a tandem head-to-tail orientation on chromosome 7. This particular gene encodes the ITIM-bearing member of the receptor pair, which functions in the inhibitory role. Alternative splicing has been observed at this locus and three variants, each encoding a distinct isoform, are described. [provided by RefSeq, Jul 2008]

PILRA: Paired receptors consist of highly related activating and inhibitory receptors and are widely involved in the regulation of the immune system. PILRA is thought to act as a cellular signaling inhibitory receptor by recruiting cytoplasmic phosphatases like PTPN6/SHP-1 and PTPN11/SHP-2 via their SH2 domains that block signal transduction through dephosphorylation of signaling molecules. Receptor for PIANP. 4 isoforms of the human protein are produced by alternative splicing.
